Target Information
Janus kinase 3 (JAK3) phosphorylates members of the STAT (signal transduction and activators of transcription) family. JAK3 is predominantly expressed in immune cells and transduces a signal in response to activation of interleukin receptors. Upon cytokine or interferon stimulation, JAKs are activated via phosphorylation at adjacent tyrosine residues, leading to STAT activation, dimerization, and nuclear translocation. Activated STAT proteins regulate transcription of numerous genes, including interferon-stimulated genes. JAK2 is required for IFNγ receptor signaling, with JAK1 acting as an amplifier. Some studies suggest that Tyk2 and JAK3 may substitute for JAK2 under certain conditions.
