BRCA1 (Breast and Ovarian Cancer Susceptibility Protein 1)
BRCA1 is a nuclear phosphoprotein involved in maintaining genomic stability and functions as a tumor suppressor. It forms part of the BASC complex (BRCA1-associated genome surveillance complex) with other tumor suppressors and DNA repair proteins. BRCA1 interacts with RNA polymerase II and histone deacetylase complexes, playing roles in transcription regulation, double-strand DNA break repair, and recombination.
Mutations in the BRCA1 gene account for about 40% of inherited breast cancers and over 80% of inherited breast and ovarian cancers. Alternative splicing affects its localization and function, with multiple transcript variants described, including disease-associated mutations. A related pseudogene is also located on chromosome 17.
