사용 전 단백질 용액을 짧게 원심분리하여 상등액만 실험에 사용하는 것이 좋습니다. 이 과정은 저장 중 형성될 수 있는 단백질 응집체를 제거하여 비특이적 배경 신호를 줄여줍니다.
Target Information
GFP (Green Fluorescent Protein)은 해파리 Aequorea victoria에서 유래된 27 kDa 단백질로, 자가 촉매적으로 형광 색소를 형성합니다.
GFP는 진핵 및 원핵 세포에서 유전자 발현 리포터나 단백질 태그로 널리 사용되며, 단백질의 위치, 이동, 세포 내 구획의 역학 연구에 활용됩니다.
야생형 GFP는 395 nm에서 흡수하고 508 nm에서 녹색 형광을 방출합니다. 추가적인 단백질, 기질, 보조 인자 없이도 형광을 나타내며, 다양한 생리학적 연구에 적합합니다.
또한 GFP는 단백질 간 상호작용 연구(yeast two-hybrid system) 및 FRET 실험에서 거리 측정에도 사용됩니다.
