Target Information
Lipolytic enzymes are essential for mobilizing fatty acids from triglyceride stores in adipose tissue. Dysfunctional lipolysis affects energy homeostasis and may contribute to obesity and insulin resistance. Hormone-sensitive lipase (HSL) was previously considered the only enzyme hydrolyzing triglycerides in mammalian adipose tissue, but adipose triglyceride lipase (ATGL) is now known to catalyze the initial step in triglyceride hydrolysis.
ATGL is highly expressed in adipose tissues of mice and humans, showing high substrate specificity for triacylglycerol and association with lipid droplets. Inhibition of ATGL decreases total adipose acyl-hydrolase activity, indicating that ATGL and HSL coordinate triglyceride catabolism in mammals.
