Product Specific Information

The 236A/E7 antibody reacts with human Foxp3 protein (FORKHEAD BOX P3, SCURFIN, JM2). Foxp3 (49–55 kDa) belongs to the forkhead/winged-helix family of transcriptional regulators and is the gene defective in scurfy (sf) mice.
Foxp3 is highly expressed in CD4+CD25+ regulatory T cells (Treg) and induces a Treg phenotype when expressed in CD4+CD25− cells.

Intracellular staining and flow cytometric analysis of human PBMCs using the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523) show staining of the CD4+CD25^bright population.

The 236A/E7 epitope differs from that of PCH101 (Product # 72-5776-40). This antibody also recognizes rhesus macaque, sooty mangabey, and cynomolgus macaque.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated for intracellular staining and flow cytometry using the Foxp3/Transcription Factor Buffer Set (Product # 00-5523-00).

Use at 5 µL (0.125 µg) per test in 100 µL final volume. Suitable for 10⁵–10⁸ cells/test.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis, expressed mainly in CD4+CD25+ regulatory T cells.
FOXP3 defects cause the scurfy phenotype in mice and IPEX syndrome (immune dysfunction, polyendocrinopathy, enteropathy, X-linked) in humans.
It may serve as a master regulatory gene and a specific marker for regulatory T cells.
In human breast and colon cancer cells, FOXP3 expression is regulated by p53 in response to DNA damage.
