Product Specific Information
The BM8 monoclonal antibody reacts with mouse F4/80 antigen (~160 kDa surface receptor), a member of the EGF-TM7 family. It contains seven EGF-like domains, seven transmembrane regions, and an intracellular domain homologous to TM7 superfamily members.
F4/80 is a well-known marker for mature macrophages but can also be expressed on peritoneal eosinophils, Langerhans cells, and certain dendritic cell subtypes. Expression begins during early myeloid development and can be upregulated by M-CSF stimulation.
Applications Reported: Flow cytometric analysis.
Applications Tested: Mouse peritoneal exudate cells, ≤2 µg/test (100 µL final volume).
Excitation: 633–647 nm; Emission: 660 nm; Laser: Red Laser.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
F4/80 antigen is a 160 kDa glycoprotein belonging to the EGF-TM7 family, sharing 68% identity with human EMR1. Expression is heterogeneous and varies with macrophage maturation and activation. It is found on tissue macrophages such as Kupffer cells, Langerhans cells, microglia, and macrophages in spleen, thymus, and bone marrow stroma.
Absent in T-cell area macrophages, F4/80 may play a role in efferent CD8⁺ regulatory T cell generation.
