Product Specific Information

The RA3-6B2 monoclonal antibody reacts with the exon A-restricted isoform of mouse CD45, a 220 kDa surface molecule. CD45R/B220 epitope is mainly expressed by the B cell lineage from early Pro-B to mature B cells. Some activated T cells, NK cell progenitors, and T cells of the lpr/lpr mutant mouse also express this antigen.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤1.0 µg/test (100 µL final volume).
Recommended to titrate for optimal performance.

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.

• Add 5 µL per sample to antibody cocktails before mixing with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor conjugates utilize Phiton™ technology, providing engineered fluorescent signatures with reduced spillover and spread.
Excitation: 552 nm; Emission: 592 nm; Laser: 561 nm (Yellow).

Note: NovaFluor Yellow 590 may non-specifically stain B cells in SJL mice. No non-specific staining observed in BALB/c or C57BL/6 strains.

Target Information

CD45R (PTPRC) is a protein tyrosine phosphatase involved in regulating cell growth, differentiation, and signaling.
It is expressed in hematopoietic cells and plays an essential role in T- and B-cell receptor signaling.
CD45 exists in multiple isoforms due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic domains.
It also contributes to integrin-mediated signal transduction and apoptosis regulation.
CD45RA is a restricted isoform expressed in specific lymphoid cell subtypes.
Dysfunction of CD45 is associated with Severe Combined Immunodeficiency and Hepatitis C.
