Product Specific Information

The RA3-6B2 monoclonal antibody reacts with the exon A-restricted isoform of mouse CD45, a 220 kDa surface molecule. The CD45R/B220 epitope is mainly expressed by B cell lineage from early Pro-B to mature B cells. Some activated T cells, lymphokine-activated killer (LAK) cells, NK progenitors, and lpr/lpr T cells also express this antigen.

• Applications Tested: Flow cytometric analysis of mouse splenocytes, ≤0.2 µg/test (in 100 µL final volume)

• 1 vial of CellBlox Plus Blocking Buffer

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology, offering engineered fluorescent signatures with minimized spillover and spread.
Excitation: 552 nm; Emission: 813 nm; Laser: 561 nm (Yellow).

Note: NovaFluor Yellow 810 may show non-specific staining of B cells in SJL mice but not in BALB/c or C57BL/6 strains.

Target Information

CD45R (PTPRC) is a protein tyrosine phosphatase regulating cell growth, differentiation, and signaling in hematopoietic cells. It plays a crucial role in T- and B-cell receptor signaling and lymphocyte development. CD45 exists in multiple isoforms due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic domains. It also modulates integrin-mediated signaling and apoptosis.
CD45RA is a specific isoform with restricted expression among lymphoid subtypes.
Associated diseases include Severe Combined Immunodeficiency (T-/B-cell negative) and Hepatitis C.
