Product Specific Information

The 2B8 monoclonal antibody reacts with mouse CD117 (c-Kit receptor), also known as Steel factor receptor or stem cell factor receptor.
This 145-kDa tyrosine kinase receptor is expressed on hematopoietic progenitor cells (Sca-1⁺ Thy1^low Lineage⁻) in mouse bone marrow.
Interaction of c-Kit with its ligand promotes proliferation and differentiation of hematopoietic progenitor cells. CD117 is also expressed by mast cells and is involved in their activation.

• Applications Tested: Flow cytometric analysis of mouse bone marrow cells (≤ 0.5 µg/test)
• Recommended Volume: 100 µL per test (10⁵–10⁸ cells/test, titrate for optimal performance)

Brilliant Violet™ 605 (BV605) is a tandem dye (emission 605 nm) for use with a violet (405 nm) laser. Ensure instrument compatibility.

Usage Notes

• Use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) when combining multiple polymer dyes to minimize non-specific interactions.
• Light sensitivity: Protect from light.
• Fixation: Samples may be stored in IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54) for up to 3 days at 4°C in the dark.
• Methanol-based fixation is not compatible with BV605.
• Excitation: 407 nm; Emission: 605 nm; Laser: Violet Laser.

Target Information

KIT (c-KIT) is a proto-oncogene encoding a type 3 transmembrane receptor for MGF (mast cell growth factor), also known as stem cell factor (SCF).
KIT regulates growth and activation of hematopoietic and non-hematopoietic cells.
Mutations in KIT are associated with GIST, mast cell disease, AML, and piebaldism.
The D816V mutation in KIT is a target in therapies for neoplastic mast cell growth.
c-Kit expression is found in hematopoietic stem cells, germ cells, mast cells, and GI tract cajal cells.
Upon SCF binding, c-Kit dimerizes, leading to receptor activation and autophosphorylation, enabling Grb2 docking and Ras/ERK signaling activation.
SCF/c-Kit also activates PI3K, PLC-γ, and JAK/STAT pathways, essential for hematopoiesis, stem cell maintenance, and gametogenesis.
