Target Information

Cholesterol transporter that mediates non-vesicular transport of cholesterol from the plasma membrane (PM) to the endoplasmic reticulum (ER). Contains unique domains for binding cholesterol and the PM, serving as a molecular bridge for cholesterol transfer from the PM to the ER. Plays a crucial role in cholesterol homeostasis and can localize to the PM based on membrane cholesterol levels. In lipid-poor conditions, localizes to the ER membrane, and in response to excess cholesterol in the PM, is recruited to ER–PM contact sites mediated by the GRAM domain. At these sites, the sterol-binding VASt/ASTER domain binds to cholesterol in the PM and facilitates its transfer to the ER. May play a role in tumor progression and autophagy regulation, being required for autophagosome biogenesis. This function in autophagy depends on its cholesterol-transfer activity. [UniProt]
