Target Information

Oncogene ERBB4 (HER4) is a member of the type I receptor protein tyrosine kinase family, which includes EGFR, ERBB2, and ERBB3. ERBB4 binds and is activated by neuregulins and other factors, inducing cellular responses such as mitogenesis and differentiation.
ErbB4 functions as a receptor for heregulin and mediates ligand-stimulated tyrosine phosphorylation. It regulates key cellular processes including metabolism, transcription, cell cycle progression, cytoskeletal rearrangement, apoptosis, and differentiation.
ErbB4 is predominantly expressed in several breast carcinoma cell lines and in normal tissues including skeletal muscle, heart, pituitary, brain, and cerebellum. High mRNA expression is observed in T47-D, MDA-MB-453, BT-474, and H3396 cell lines, while lower expression is seen in MCF-7, MDA-MB-330, and MDA-MB-361. Expression is low or absent in MDA-MB-231, MDA-MB-157, MDA-MB-468, and SKBR-3. Elevated ErbB4 levels in certain tumor cell lines suggest a possible role in malignancy.
