Product Specific Information

The eBioC9B7W monoclonal antibody recognizes mouse CD223 (LAG-3, LAG3) expressed by activated alpha/beta-TCR-bearing T cells, subsets of gamma/delta-TCR T cells, and NK cells.
CD223 is a 70 kDa type I transmembrane protein similar to CD4. Mouse CD223 can be proteolytically cleaved, producing a 54 kDa extracellular fragment and a 16 kDa intracellular fragment. The soluble form may regulate T-cell activation and homeostatic proliferation.

This antibody has been tested by flow cytometric analysis of stimulated mouse splenocytes at ≤1 µg/test (100 µL final volume). Cell count per test should be optimized (10⁵–10⁸ cells/test).

APC-eFluor® 780 emits at 780 nm and is excited with a 633 nm red laser. Ensure instrument compatibility.
Light-sensitive: protect vial and stained samples from light.

Fixation: Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-822-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54). Performance after fixation should be verified empirically.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

LAG-3 is a 70-kDa surface glycoprotein of the Ig superfamily, homologous to CD4. It binds MHC class II with higher affinity than CD4 and negatively regulates T-cell activation and proliferation. Expressed on activated T cells (including regulatory T cells) and NK cells. CD8+ T cells generally express higher levels than CD4+ T cells. Coexpression with CD49b identifies Type 1 regulatory T cells (Tr1 cells) in humans and mice.
