Product Specific Information
The 14D3 monoclonal antibody reacts with human CD152 (CTLA-4), a protein structurally similar to CD28. It is expressed on activated T cells (and sometimes B cells) and binds CD80 (B7-1) and CD86 (B7-2) with higher affinity than CD28.
CTLA-4 and CD28 deliver opposing signals: CD28 stimulates T cell activation, while CTLA-4 inhibits IL-2 secretion and proliferation. Most CTLA-4 is localized intracellularly due to ER retention motifs.
The 14D3 antibody also recognizes rhesus monkey and exhibits inhibitor activity.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated using the Intracellular Fixation and Permeabilization Buffer Set (Product #88-8824-00) following BestProtocols® Protocol A.
Use 5 µL (0.06 µg) per test in a final volume of 100 µL. Cell count may range from 10⁵–10⁸ cells/test.
Due to intracellular localization, both surface and intracellular CTLA-4 should be assessed for complete detection.
Fluorochrome Properties:
- Excitation: 633–647 nm
- Emission: 780 nm
- Laser: Red Laser
- Light Sensitivity: Protect from light (photo-oxidation sensitive)
Fixation: Samples can be stored up to 3 days at 4°C in the dark using:
- IC Fixation Buffer (Product #00-8222), or
- 1-step Fix/Lyse Solution (Product #00-5333)
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor mainly expressed on CD4⁺ T cells.
It binds CD80 and CD86 with higher affinity than CD28, but unlike CD28, CTLA4 inhibits T cell activation and function.
Intracellular CTLA4 is also present in regulatory T cells, contributing to their immunoregulatory role.
