Product Specific Information

MA3-062 detects spectrin from mouse, rat, and human erythrocytes, brain, and muscle cells.
This antibody specifically detects two alternatively spliced forms of spectrin:

• Beta-1 epsilon-1 (erythrocytes)
• Beta-1 epsilon-2 (nerve and striated muscle cells)

It does not cross-react with alpha-2 spectrin or fodrin subunits.
MA3-062 has been successfully used in Western blot, immunohistochemistry (paraffin), and immunofluorescence.
By Western blot, it detects a ~246 kDa β-1 spectrin protein in rat skeletal muscle homogenate.
Immunofluorescence staining shows intense sarcolemma staining in rat skeletal muscle fibers.

Fixation Note:
In immunofluorescence, aldehyde fixatives can compromise antigen integrity.
If using paraformaldehyde, exposure should be limited to ≤5 minutes with ≤2% solution.

Antigen used: Purified human erythrocyte beta-1 spectrin.

Target Information

Spectrin (Sp) is the most abundant erythrocyte membrane skeleton protein, maintaining cell shape and flexibility.
Erythrocyte spectrin is a heterodimer of a 280 kDa α-subunit and a 246 kDa β-subunit, forming a 100 nm rod-like structure.
Spectrin in other tissues may consist of homologous α and β subunits (fodrin).
The nomenclature designates erythrocyte subunits as α-1 and β-1, and fodrin subunits as α-2 and β-2.
Alternatively spliced forms are labeled ε-1, ε-2, etc. (e.g., β-1 ε-1).
