Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. p53 upregulates genes such as p21waf1, contributing to G1 or G2/M arrest. Structurally, p53 consists of:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerisation domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is crucial for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 functions as a tumor suppressor, preventing proliferation of damaged cells, and is linked to aging, oxidative stress, and cognitive function. Mutations in TP53 result in loss of tumor suppressor activity and are common in various human cancers, including hereditary cancer syndromes such as Li-Fraumeni syndrome.
