Product Specific Information

Description:
The H57-597 monoclonal antibody reacts with the beta chain of mouse TCR. TCR beta is expressed by thymocytes in a developmentally regulated manner and by most peripheral T cells. Crosslinking of the TCR complex with H57-597 can induce T cell activation, proliferation, or apoptosis depending on assay conditions. It serves as a phenotypic marker for TCR beta-expressing T cells.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.125 µg per test (in 100 µL final volume). Cell number per test may range from 10⁵ to 10⁸ cells. Antibody titration is recommended for optimal performance.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended detection filter: 610/20 bandpass. Ensure the instrument supports this fluorochrome.

Usage Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples may be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark, with minimal loss of brightness or FRET efficiency.

Target Information

T cell receptors (TCRs) enable T cells to recognize peptides presented by MHC class II molecules, essential for adaptive immune responses. Recognition of self-peptides is linked to autoimmune diseases, while mutant self-peptides are associated with tumors. Superantigen engagement of TCRs can lead to toxic shock syndrome. Autoantibodies against TCR V beta segments have been found in rheumatoid arthritis and lupus patients, potentially modulating inflammatory reactions.
Most human T cells express TCR alpha-beta with CD4 or CD8 (single positive). Some are double negative (DN), and increased DN T cells are observed in autoimmune or immunodeficiency disorders. Gamma-delta T cells, found mainly in epithelium, show distinct antigen recognition and may have antitumor and immunoregulatory roles.
