Product Specific Information

The H74 monoclonal antibody reacts with human B7-S1 (also known as B7-H4 or B7x). Cross-reactivity with other proteins has not been determined. B7S1 is a member of the B7 family, involved in the costimulatory regulation of T cells, potentially influenced by B cell activation status. BTLA has been suggested as a counter receptor, though further validation is needed.

• Applications Reported: Suitable for flow cytometric analysis.
• Applications Tested: Pre-diluted and tested by flow cytometry on B7-H4-transfected human cells. Recommended use: 5 µL (0.25 µg) per test (100 µL total volume, 10⁵–10⁸ cells/test).

Super Bright 600 Characteristics:

• Tandem dye excited by 405 nm violet laser; emits at 600 nm (recommended 610/20 bandpass filter).
• Use Super Bright Complete Staining Buffer (Product # SB-4401) when combining multiple Super Bright conjugates to minimize non-specific interactions.
• Light-sensitive: protect from light during storage and staining.
• Fixation: Compatible with IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57); samples stable up to 3 days at 4°C in dark.
• Excitation: 405 nm / Emission: 600 nm / Laser: Violet.

Target Information

B7-H4 (B7x, VTCN1) is a costimulatory protein functioning as a negative regulator of T-cell mediated immunity. It delivers inhibitory signals to T-cells, suppressing proliferation, cell cycle progression, and IL-2 production.

• Expressed on activated T-cells, B-cells, monocytes, and dendritic cells.
• Aberrant expression is linked to lung, breast, and ovarian cancers.
• Overexpression may enable tumor immune evasion and progression.
• Potential biomarker for early detection of ovarian cancer and renal cell carcinoma (RCC).
