Target Information
Thymidylate Synthase (TS)는 deoxyuridine monophosphate (dUMP)를 deoxythymidine monophosphate (dTMP)로 전환시켜 DNA 생합성에 필수적인 효소입니다. TS는 항암제인 fluoropyrimidines 계열 약물의 주요 표적이며, 이 약물들은 고형암 치료에 널리 사용됩니다. 5-FU 및 fluorodeoxyuridine은 종양 세포 내에서 FdUMP로 전환되어, folate 보조인자 5,10-methylenetetrahydrofolate 존재 하에 TS를 불활성화하는 삼중 공유 복합체를 형성합니다. TS 단백질의 발현은 인간의 대장암, 위암, 두경부암, 유방암 등에서 5-FU 반응성과 관련이 있습니다.
