Target Information
GAPDH (Glyceraldehyde-3-phosphate dehydrogenase)는 해당과정(glycolysis)에 관여하는 촉매 효소로, 37 kDa의 동일한 소단위체 4개로 구성된 사량체 형태로 존재합니다.
이 효소는 NADPH 존재하에 1,3-bisphosphoglycerate를 glyceraldehyde-3-phosphate로 환원시키는 반응을 촉매합니다.
해당과정 이외에도 GAPDH는 다음과 같은 다양한 세포 기능에 관여합니다.
- DNA 복제 및 수선
- 핵 RNA 수출
- 세포막 융합 및 미세소관 결합
- 세포자멸사(apoptosis) 및 신경퇴행성 질환 관련 유전자 발현 조절
또한 GAPDH는 Alzheimer’s disease의 원인 단백질인 amyloid precursor protein 및 Huntington’s disease 관련 단백질 Huntingtin과 결합하는 것으로 보고되었습니다.
GAPDH는 대부분의 조직에서 높은 수준으로 발현되므로 Western blot에서 단백질 로딩 컨트롤로 널리 사용됩니다.
