Target Information
CD33 is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit protein tyrosine phosphatases SHP-1 and SHP-2, contributing to signal regulation and receptor removal via ubiquitination.
CD33 is expressed on myelomonocytic lineage cells and binds sialic acid residues on cell surfaces. It is a known therapeutic target for acute myeloid leukemia (AML) and is found on granulocyte and macrophage precursors in the bone marrow, but not on pluripotent stem cells.
Additionally, CD33 serves as a marker for peripheral monocytes and is useful for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
