Target Information
KIT (c-KIT)은 MGF(mast cell growth factor, stem cell factor) 수용체로 작용하는 프로토온코진이며, type 3 transmembrane receptor입니다. KIT은 처음에 feline sarcoma viral oncogene v-kit의 세포 상동체로 확인되었습니다.
KIT과 그 리간드는 다양한 조혈 및 비조혈 세포의 성장과 활성화를 조절합니다. KIT의 돌연변이는 위장관 기질 종양(GIST), 비만세포 질환, 급성 골수성 백혈병, piebaldism 등과 관련이 있습니다.
특히 D816V 활성 루프 돌연변이에 의한 KIT 수용체 티로신키나제의 비정상적 활성화는 비정상 비만세포 성장 억제를 위한 치료 전략의 핵심으로 연구되고 있습니다.
c-Kit은 조혈 줄기세포, 생식세포, 비만세포 및 위장관의 Cajal 세포에서 발현되며, SCF(stem cell factor) 결합 시 이합체화되어 자가 인산화 및 다양한 신호경로(Grb2/Ras/ERK, PI3K, PLC-γ, JAK/STAT 등)를 활성화합니다.
이러한 경로는 조혈, 줄기세포 유지, 생식세포 형성에 필수적입니다.
