Product Specific Information

Description:
Monoclonal antibody 1A8-Ly6g reacts with mouse Ly-6G (Gr-1), a 25-kDa GPI protein expressed exclusively by neutrophils. It plays a role in neutrophil infiltration, recruitment, and migration, and associates with β2 integrins (CD11a, CD11b). The ligand for Ly-6G is currently unknown.

Unlike RB6-8C5, the 1A8-Ly6g antibody binds specifically to Ly-6G on neutrophils and has been used for neutrophil depletion with reported antagonist activity.

Applications Reported:

• Flow cytometric analysis of mouse bone marrow cells.

Applications Tested:

• Use ≤ 1.0 µg/test (100 µL final volume).
• Optimal cell number: 10⁵–10⁸ cells/test.
• Titrate for optimal performance.

Fluorochrome Information:
Brilliant Ultra Violet™ 395 (BUV395) emits at 395 nm and is designed for cytometers with a 355 nm UV laser. Ensure instrument compatibility.

When using multiple polymer dye-conjugated antibodies (Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use:

Target Information

Ly6G (Lymphocyte antigen 6 complex locus G6D):
A component of the myeloid differentiation antigen Gr-1, along with Ly6C.

• Molecular weight: ~21–25 kDa
• Expressed by myeloid-derived cells in bone marrow
• Marker for peripheral neutrophils, monocytes, and granulocytes
• Expression correlates with granulocyte differentiation and maturation
• Involved in anti-tumor response and may inhibit local immune responses
