Product Specific Information

The SIDI8BEE monoclonal antibody reacts with human and rhesus macaque CD8 beta. The CD8β chain associates with the CD8α chain to form the CD8αβ heterodimer expressed on thymocytes and peripheral cytotoxic T cells. CD8 receptor complex binds to MHC class I and plays a key role in T cell development and activation.

SIDI8BEE detects CD8β in both CD8αβ heterodimer and CD8ββ homodimer. Cross-reactive with human and rhesus macaque CD8β; other primates not tested.

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on human peripheral blood cells at 5 µL (0.2 µg) per test in 100 µL final volume. Recommended cell number: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell suspension (10³–10⁸ cells).

Target Information

CD8B is a cell surface glycoprotein expressed on cytotoxic T lymphocytes, functioning as a coreceptor with the T-cell receptor to recognize antigens presented by MHC class I molecules. The CD8 complex can form as αα homodimers or αβ heterodimers. This gene encodes CD8β chain isoforms, with multiple alternatively spliced variants and a pseudogene located on chromosome 2.
