Product Specific Information
The 14D3 monoclonal antibody reacts with human CD152 (CTLA-4), a protein structurally related to CD28. CTLA-4 is expressed on activated T cells and binds CD80 (B7-1) and CD86 (B7-2) with higher affinity than CD28. While CD28 acts as a costimulator, CTLA-4 inhibits T cell activation by reducing IL-2 secretion and proliferation. A significant portion of CTLA-4 is localized intracellularly due to ER retention motifs.
The 14D3 antibody also recognizes rhesus monkey CTLA-4 and exhibits inhibitory activity.
Applications Reported: Flow cytometry, Western blot, and in vitro inhibition studies.

Applications Tested:
Validated for intracellular staining and flow cytometric analysis of PHA-stimulated human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
Recommended usage: ≤0.25 µg/test (100 µL final volume). Optimal antibody concentration should be titrated per assay.
Due to intracellular localization, complete detection of CTLA-4 may require assessment of both surface and intracellular expression.

Target Information
CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor primarily expressed on CD4+ T cells. It binds CD80 and CD86 with higher affinity than CD28, acting as an inhibitory receptor that downregulates T cell activation. Intracellular CTLA4 is also found in regulatory T cells, contributing to immune regulation.
