Product Specific Information

The monoclonal antibody P1H12 recognizes CD146 (also known as MUC18, s-endo, Endo-CAM, and Mel-CAM), a member of the immunoglobulin superfamily. CD146 is expressed on endothelial cells, bone marrow fibroblasts, and certain tumors (notably melanoma). It is also found on mesenchymal stromal and endometrial stromal cells.

Expression of CD146 on circulating blood cells is limited to a subset of T cells rather than endothelial cells, as markers such as CD31 and CD51/61 are negative. CD146 is expressed on activated lymphocytes and is heavily glycosylated, with over 50% of its mass derived from carbohydrates.

The P1H12 antibody cross-reacts with mouse, rabbit, and canine, but not rat.

Applications Reported: Flow cytometry, immunoprecipitation, immunoblotting (WB under nonreducing conditions), and immunohistology of frozen tissue sections.
Applications Tested: Validated by flow cytometric analysis of HUVEC cells. Recommended ≤0.25 µg/test (per 100 µL, 10⁵–10⁸ cells/test). Optimize concentration as needed.

Target Information

CD146 (MCAM) is an integral membrane glycoprotein of the immunoglobulin superfamily, expressed at cell-cell junctions in endothelial cells and other cell types. It functions as a calcium-independent adhesion molecule and may act in signal transduction via Fyn kinase, influencing actin cytoskeleton assembly.

CD146 contributes to endothelial cohesion and melanoma cell interaction with vascular elements, facilitating tumor spread. It may also play roles in neural crest cell adhesion during embryonic development and act as a receptor triggering tyrosine phosphorylation of FYN and PTK2, with transient intracellular calcium increases.
