Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. p53 activation leads to transcription of genes such as p21waf1, contributing to G1 or G2/M arrest. Structurally, p53 includes an N-terminal transactivation domain, central DNA-binding domain, oligomerization domain, and C-terminal regulatory domain. Phosphorylation at Ser15 is crucial for activation and stabilization, while Ser392 phosphorylation is associated with tumor formation. p53 plays key roles in preventing proliferation of damaged cells, aging, oxidative stress response, and cognitive processes like long-term potentiation. It is expressed at low levels in normal cells but elevated in transformed cells, contributing to malignancy. Mutations in TP53 disrupt DNA binding and tumor suppressor activity, frequently occurring in human cancers and inherited cancer syndromes such as Li-Fraumeni syndrome.
