Target Information

TAK1 (MAP3K7) is a serine/threonine kinase involved in multiple signaling pathways. Initially identified as a TGF-β-activated kinase, TAK1 interacts with TGF-β receptor and TRAF6 to modulate activation of JNK and p38. It also participates in Wnt, BMP, and activin signaling, as well as responses to thyroid hormone, osmotic stress, and endothelin.

TAK1 regulates wound healing by promoting von Hippel-Lindau tumor suppressor expression and repressing PDGF-B, integrin β1, and β5. It is crucial for IKK activation in pathways including IL-1, IL-6, IL-18, TNF, CD40, TLR, and RIG-I. Activation involves TAB2/3 binding and ubiquitination, followed by TAK1 autophosphorylation and phosphorylation of IKK-β. TAK1 also represses human telomerase reverse transcriptase, implicating it in cell lifespan regulation.
