Product Specific Information

The monoclonal antibody QCH6A7 reacts with mouse CD20, a B cell-specific 32 kDa membrane protein expressed from late pro-B cells through memory B cells. CD20 is not found on hematopoietic stem cells, early pro-B cells, or mature plasma cells. It regulates intracellular calcium and plays a role in B cell proliferation.

CD20 expression begins after CD19 induction and increases during maturation. Unlike other B cell antigens, CD20 is not shed or internalized upon antibody binding. Monoclonal antibodies against CD20 can deplete B cells and are used as immunotherapeutic targets in B cell lymphomas and autoimmune diseases such as rheumatoid arthritis and type I diabetes.

CD20 has four transmembrane domains and a 44-amino-acid extracellular domain containing the QCH6A7 epitope. Staining with clone QCH6A7 provides improved staining intensity compared to clone AISB12 in flow cytometric assays.

Target Information

CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and most malignant B cells, as well as a subset of T lymphocytes and follicular dendritic cells. It regulates calcium conductance, cell cycle progression, and B-cell proliferation.

CD20 is associated with lipid rafts, and its distribution depends on extracellular signals and BCR aggregation. After receptor ligation, CD20 remains at the cell surface while BCR is internalized. CD20 is a key therapeutic target for B-cell depletion in malignancies, as it remains membrane-bound after antibody treatment.
