Target Information

BCL-2 is a key regulator of apoptosis that functions to either inhibit or promote cell death. Members of the BCL-2 family can dimerize to modulate apoptosis. For example, Bag1 forms a heterodimer with BCL-2, enhancing its anti-apoptotic effect. Bax and Bak are critical for cytochrome c release from mitochondria, initiating apoptosis. Bax exerts a pro-apoptotic effect, while constitutive expression of BCL-2, such as from translocation to the Ig heavy chain locus, is associated with follicular lymphoma. In most follicular lymphomas, neoplastic germinal centers express high levels of BCL-2 alpha protein, whereas normal or hyperplastic germinal centers are negative. Two transcript variants of BCL-2 differ in their C-terminal ends. Overexpression of BCL-2 has been linked to cancers such as B-cell lymphoma and prostate cancer.
