Target Information
The NF-κB/Rel transcription factors reside in the cytosol in an inactive state, complexed with inhibitory IκB proteins. Activation of NF-κB typically occurs through phosphorylation-induced, proteasome-mediated degradation of IκB.
A key regulatory step involves activation of the IκB kinase (IKK) complex, composed of catalytic subunits IKKα and IKKβ and regulatory subunit IKKγ. Activation depends on phosphorylation at Ser177 and Ser181 in IKKβ (Ser176 and Ser180 in IKKα), leading to conformational changes and kinase activation.
