Target Information
STAT3는 STAT (signal transducers and activators of transcription) 단백질 계열에 속하며, 수용체 관련 키나아제에 의해 인산화되어 핵으로 이동한 후 전사인자로 작용합니다. STAT3는 사이토카인 및 성장인자에 반응하여 전사 활성화를 유도하며, CREB-binding protein과 같은 코액티베이터가 필요합니다.
STAT3는 Interferon-α, Interferon-γ, EGF, PDGF, IL6 등에 의해 활성화될 수 있으며, JAK1 및 JAK2에 의해 Tyr705 잔기의 인산화가 STAT3 이합체 형성, 핵 이동, DNA 결합 활성에 필수적입니다.
인간 STAT3 유전자는 염색체 17q에 위치하며, IFN-α에 의해 활성화되지만 IFN-β에는 반응하지 않습니다. STAT3는 세포 성장, 분화, 아폽토시스 등 다양한 세포 과정에 관여하며, Rac1 GTPase 및 PIAS3 단백질에 의해 조절됩니다.
세 가지 대체 스플라이싱 전사 변이가 보고되어 있으며, STAT3 유전자 결실은 생쥐 배아 단계에서 치명적입니다.
