Product Specific Information

The RPA-T8 monoclonal antibody reacts with the human CD8a molecule (~32–34 kDa), expressed as a CD8α/β heterodimer or CD8α/α homodimer. CD8a is found on most thymocytes and subsets of mature T and NK cells. It binds MHC class I and associates with tyrosine kinase p56lck, playing a role in T-cell development and activation.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Pre-diluted and validated for flow cytometry of human peripheral blood cells (5 µL/test; 0.25 µg/test).
    A test corresponds to staining a 100 µL cell sample (10⁵–10⁸ cells/test).
  • Fluorochrome Info: Brilliant Violet™ 711 (BV711) emits at 713 nm; suitable for cytometers with a 405 nm violet laser.
  • Buffer Recommendation: Use Super Bright Complete Staining Buffer (#SB-4401-42) or Brilliant Stain Buffer™ (#00-4409-75) when combining polymer dyes to minimize non-specific interactions.
  • Light Sensitivity: Protect from light to prevent photo-induced oxidation.
  • Fixation: Compatible with IC Fixation Buffer (#00-8222-49) or 1-step Fix/Lyse Solution (#00-5333-54) for up to 3 days at 4°C.
    Not compatible with methanol-based fixation.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein in the immunoglobulin receptor family. It functions as a co-receptor with TCR for MHC class I molecules, mediating signal transduction and T cell activation. CD8 is expressed mainly on cytotoxic T cells, NK cells, monocytes, and dendritic cells. Upon MHC-I/peptide binding, CD8 recruits Lck kinase, leading to cytokine production and cytotoxic T lymphocyte (CTL) activation, crucial for pathogen and tumor clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
