Target Information

Matrix metalloproteinases (MMPs) are proteolytic enzymes capable of degrading connective tissue components. They share a common activation mechanism, a conserved metal-binding active site, and are inhibited by tissue inhibitors of metalloproteinases (TIMPs). MMPs and TIMPs play important roles in angiogenesis regulation.

MMP-2 is synthesized as a 631-amino acid proenzyme, activated by cleavage of the first 80 amino acids, and includes propeptide, catalytic, and hemopexin domains. MMPs can degrade extracellular matrix components such as collagens, fibronectin, laminin, and basement membrane glycoproteins. Structurally, they possess a signal peptide, propeptide, and catalytic domain with a conserved zinc-binding site. Functionally, MMP-2 is involved in tissue remodeling. Mutations in the MMP-2 gene are associated with Winchester syndrome and Nodulosis-Arthropathy-Osteolysis (NAO) syndrome. Two transcript variants encoding different isoforms of MMP-2 have been identified.
