Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 activation induces expression of genes such as p21waf1, regulating G1 or G2/M arrest.

Structurally, p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is critical for activation and stabilization. Phosphorylation at Ser392 correlates with tumor formation. p53 plays a key role in preventing proliferation of damaged cells and acts as a tumor suppressor. Mutations in TP53 disrupt DNA binding and tumor suppression, contributing to cancer development. Germline TP53 mutations are associated with Li-Fraumeni syndrome.
