Target Information

Matrix metalloproteinases (MMPs) are proteolytic enzymes capable of degrading connective tissue components. They share a common activation mechanism and a conserved metal-binding active site sequence, and are inhibited by tissue inhibitors of metalloproteinases (TIMPs). MMPs and TIMPs play key roles in angiogenesis regulation.

MMP2 is synthesized as a 631-amino-acid proenzyme, activated by cleavage of the first 80 amino acids, and contains propeptide, catalytic, and hemopexin domains. MMPs collectively degrade extracellular matrix components such as collagens, fibronectin, laminin, and basement membrane glycoproteins. Structurally, MMPs have a signal peptide, a propeptide, and a catalytic domain with a conserved zinc-binding site. Functionally, MMP2 is involved in tissue remodeling. Mutations in the MMP2 gene are associated with Winchester syndrome and Nodulosis-Arthropathy-Osteolysis (NAO) syndrome. Two transcript variants encoding different isoforms of MMP2 have been identified.
