Product Specific Information

This 13F12F2 monoclonal antibody reacts with human killer cell lectin-like receptor subfamily G, member 1 (KLRG1), a type II transmembrane inhibitory receptor of the C-type lectin superfamily containing an ITIM domain. It is expressed on subsets of γδ T cells, NK (CD56dim), CD8+ and CD4+ T cells, mainly in effector/memory phenotype cells that are short-lived but capable of immediate effector function.

Cadherin-E, -N, and -R are ligands for KLRG1, and their interaction inhibits cytolytic activity and proliferation. The percentage of KLRG1-positive cells varies depending on antigen exposure.
The clones 13F12F2 and 13A2 recognize a similar epitope based on cross-blocking studies.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of human peripheral blood cells; use at 5 µL (0.25 µg) per test.
A test corresponds to staining a cell sample (10⁵–10⁸ cells/test) in 100 µL final volume.
Excitation: 488–561 nm; Emission: 578 nm; Laser: Blue, Green, Yellow-Green.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

KLRG1 (Killer cell lectin-like receptor G1, MAFA, 2F1-Ag) is a homodimeric receptor of the lectin-like type 2 transmembrane family containing ITIM motifs in its cytoplasmic domain. These motifs interact with SH2 domains of phosphatases such as SHP-1. KLRG1 acts as an inhibitory receptor expressed on NK cells and certain T cells, involved in tumor and virus-infected cell lysis and immune regulation.
