Target Information
STAT3는 STAT(signal transducers and activators of transcription) 단백질 계열에 속하며, 수용체 관련 키나아제에 의해 인산화되어 핵으로 이동한 후 전사 인자로 작용합니다. STAT3의 전사 활성화에는 CREB-binding protein과 같은 코액티베이터가 필요합니다.
STAT3는 Interferon-alpha, Interferon-gamma, EGF, PDGF, IL6 등에 의해 활성화될 수 있으며, Tyr705 부위의 인산화는 JAK1 및 JAK2에 의해 매개되어 STAT3의 이합체 형성, 핵 이동, DNA 결합에 필수적입니다.
인간 STAT3 유전자는 염색체 17q에 위치하며, IFN-alpha에 의해 활성화되지만 IFN-beta에는 반응하지 않습니다. STAT3는 다양한 세포 자극에 대한 유전자 발현을 매개하여 세포 성장, 아폽토시스 등 여러 세포 과정에 중요한 역할을 합니다.
Rac1은 STAT3의 활성을 조절하고, PIAS3는 STAT3의 특이적 억제 단백질로 알려져 있습니다. STAT3는 3가지 상이한 isoform을 암호화하는 대체 스플라이싱 전사 변이를 가지며, STAT3 유전자 결손 마우스는 배아 초기 단계에서 치명적입니다.
