Target Information
STAT3는 STAT (signal transducers and activators of transcription) 단백질 계열에 속하며, 수용체 관련 키나아제에 의해 인산화되어 핵으로 이동하고, 사이토카인 및 성장 인자 자극에 반응하는 전사 인자로 작용합니다. CREB-binding protein과 같은 공동활성화 인자가 STAT3의 전사 활성화에 필요합니다.
STAT3는 Interferon-α, Interferon-γ, EGF, PDGF 및 IL6에 의해 활성화될 수 있습니다. Tyrosine 705의 인산화는 JAK1 및 JAK2에 의해 매개되며, STAT3 이합체 형성, 핵 이동 및 DNA 결합에 필수적입니다.
STAT3 유전자는 인간 17번 염색체 q arm에 위치하며, IFN-α에 의해 활성화되지만 IFN-β에는 반응하지 않습니다. STAT3와 관련된 전사 인자로는 c-Jun 및 CREB가 있으며, 세포 성장과 세포사 등 다양한 세포 과정에서 핵심 역할을 합니다. Rac1 GTPase는 STAT3의 활성을 조절하며, PIAS3 단백질은 STAT3의 특이적 억제 인자로 알려져 있습니다.
STAT3에는 서로 다른 isoform을 암호화하는 세 가지 대체 스플라이싱 전사 변형이 존재하며, STAT3 유전자 결손 마우스는 초기 배아 단계에서 치명적입니다.
