Target Information
Adhesion to the extracellular matrix regulates cell survival through both integrin engagement and proper cell spreading. Anoikis is the apoptosis mechanism induced by integrin detachment. Amino-terminal enhancer of split (AES) is a member of the Groucho/transducin-like enhancer of split (TLE) family of transcriptional regulators, functioning as co-repressors involved in neurogenesis, segmentation, and sex determination. AES forms a complex with Bit1 (Bcl-2 inhibitor of transcription 1), a mitochondrial protein released into the cytoplasm during apoptosis. This complex suppresses survival-promoting gene transcription controlled by the TLE family. Apoptosis induced by AES and Bit1 can be inhibited when cells attach to fibronectin via the α5β1 integrin, suggesting that the Bit1–AES pathway contributing to anoikis is regulated by integrins, particularly α5β1.
