Target Information
Tau는 신경세포의 축삭에서 주로 발견되는 미세소관 결합 단백질로, 튜불린 중합을 촉진하고 미세소관을 안정화시키는 역할을 합니다.
C-말단은 축삭 미세소관과 결합하고 N-말단은 신경 세포막 구성요소와 결합하여 Tau가 두 구조 사이의 연결 단백질로 기능함을 시사합니다.
TAU/MAPT의 위치에 따라 축삭 극성이 결정되며, 짧은 아이소폼은 세포골격의 유연성을, 긴 아이소폼은 안정성을 담당합니다.
과인산화된 Tau는 Alzheimer’s disease(AD) 뇌에서 신경섬유 엉킴(neurofibrillary tangles)의 주요 구성 요소이며, 미세소관 결합 기능을 저해하여 신경퇴행을 유발합니다.
Tau 인산화에는 GSK-3β, PKA, cdk5, casein kinase II 등이 관여하며, Pick’s disease, frontotemporal dementia, corticobasal degeneration, progressive supranuclear palsy 등 다양한 신경계 질환에서 발견됩니다.
