Target Information
MDM2 is a ubiquitin ligase for p53 and plays a central role in the regulation of p53 stability. It is located on chromosome 12q. Akt-mediated phosphorylation of MDM2 at Ser166 and Ser186 enhances its interaction with p300, promoting MDM2-mediated ubiquitination and degradation of p53. Phosphorylation also prevents MDM2 binding to p19ARF, further increasing p53 degradation. Overexpression of MDM2 can inactivate p53, reducing its tumor suppressor function. MDM2 also interacts with retinoblastoma 1 and ribosomal protein L5, influencing cell cycle, apoptosis, and tumorigenesis. It is overexpressed in various human cancers, including soft tissue carcinoma and breast cancer. More than 40 alternatively spliced transcript variants of MDM2 have been identified in tumor and normal tissues.
