Specificity Comments

PU.1 is a member of the ETS family of transcription factors and is important for normal B-cell development. It is expressed in the myeloid lineage and in immature as well as mature B-lymphocytes, except plasma cells. PU.1 is expressed in germinal center and mantle B-cells, and various lymphomas are positive for this marker. It is essential during early B-cell differentiation, and absence of PU.1 results in a total block of B-cell development at the pre-pro stage. PU.1 regulates the expression of immunoglobulin and other genes important for B-cell development and plays a key role in normal myeloid differentiation.

Target Information

This gene encodes an ETS-domain transcription factor that activates gene expression during myeloid and B-lymphoid cell development. The nuclear protein binds to a purine-rich sequence known as the PU-box near promoters of target genes and regulates their expression in coordination with other transcription factors and cofactors. It can also regulate alternative splicing of target genes. Multiple transcript variants encoding different isoforms have been identified.
