Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to signaling pathways, regulating cell death and stress responses. TRAF proteins share a conserved domain enabling receptor and TRAF interactions, activating distinct signaling cascades. TRAF2 and TRAF3 bind to CD40, a member of the NF receptor superfamily, and are essential for NF-kappa-B activation in mouse B lymphocytes. TRAF2, together with TRAF6, is required for CD40 signaling in nonhematopoietic cells and also interacts with the lymphotoxin-beta receptor (LTbetaR) along with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
