Target Information
The human UNG gene encodes both nuclear (UNG2) and mitochondrial (UNG1) forms of uracil-DNA glycosylase. These forms result from alternative splicing and two differentially regulated promoters (PA and PB).
UNG1 and UNG2 cDNAs are similar in size (2061 and 2058 bp) and thus not resolved as distinct species in gel electrophoresis.
Nuclear UNG2 differs from mitochondrial UNG1 by 44 amino acids in the N-terminal sequence, which is not required for catalytic activity.
UNG gene products repair mutagenic U:G mispairs caused by cytosine deamination, with UNG2 removing misincorporated dUMP residues.
Expression levels of UNG1 and UNG2 vary by cell and tissue type and are cell cycle regulated. Generally, UNG expression and activity are higher in proliferating cells and tissues.
