Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events such as the establishment and maintenance of pregnancy.
PR exists as a 94 kDa protein (Form A) or a 120 kDa protein (Form B) due to alternative translation initiation. PR-B is transcriptionally active, maintaining endometrial function and pregnancy, while PR-A lacks 165 amino acids at the N-terminus, exposing an inhibitory domain that represses steroid hormone transcriptional activity.
In its inactive state, PgR forms a multiprotein complex with heat shock proteins and immunophilins. Upon progesterone binding, conformational changes allow receptor dimerization and binding to progesterone response elements (PRE), activating transcription.
Null mutations in the PGR gene can lead to reproductive abnormalities.
