Product Specific Information
Description:
The V3GYWCH monoclonal antibody reacts with human and mouse interferon regulatory factor 8 (IRF8; ICSBP). IRF8 is a 50 kDa transcription factor critical for dendritic cell (DC) development, particularly plasmacytoid DC (pDC) and CD8a+ DC differentiation. IRF8-/- mice lack both pDC and CD8a+ DC populations, while CD8a- DCs remain but fail to mature properly. Retroviral transduction of IRF8 restores pDC development in deficient mice.

Reported for use in flow cytometric analysis.
Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤0.25 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimal titration recommended.
Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.
Fixation:
Samples can be stored in:
- IC Fixation Buffer (Product # 00-8222-49)
- 1-step Fix/Lyse Solution (Product # 00-5333-57)
for up to 3 days at 4°C in the dark with minimal impact on brightness.
Excitation/Emission:
488–561 nm excitation; 775 nm emission.
Compatible Lasers: Blue, Green, Yellow-Green.
Target Information
IRF8 (interferon regulatory factor 8) acts as a transcriptional activator or repressor, binding to the upstream regulatory regions of type I IFN and MHC class I genes. It regulates immune system cells negatively and is essential for CD8+ dendritic cell differentiation through complex formation with BATF-JUNB heterodimer. Mutations in IRF8 can cause immunodeficiency 32A and 32B.
