Target Information

JMY acts as both a nuclear p53/TP53 cofactor and a cytoplasmic regulator of actin dynamics depending on conditions.
In the nucleus, it functions as a cofactor that enhances the p53/TP53 response via interaction with p300/EP300, increasing p53/TP53-dependent transcription and apoptosis. This suggests an important role in p53/TP53 stress responses such as DNA damage.
In the cytoplasm, JMY acts as a nucleation-promoting factor for both branched and unbranched actin filaments, activating the Arp2/3 complex to induce branched actin filament networks. It can also catalyze actin polymerization in the absence of Arp2/3, forming unbranched filaments.
JMY contributes to cell motility by controlling actin dynamics and may facilitate rapid formation of branched actin networks by nucleating new mother filaments and subsequently activating Arp2/3 to branch off these filaments.
Its dual functions as a p53/TP53 cofactor and actin activator are regulated by subcellular localization.
