• Positive Control: Gastrointestinal stromal tumor (GIST) or testicular germ cell tumor. Melanocytes in the basal layer of the epidermis and mast cells in the dermis of normal skin.

Target Information

KIT (c-KIT) is a proto-oncogene and a type 3 transmembrane receptor for mast cell growth factor (MGF, also known as stem cell factor). KIT was first identified as the cellular homolog of the feline sarcoma viral oncogene v-kit. Together with its ligand, KIT regulates growth and activation of various hematopoietic and non-hematopoietic cells.

Mutations in KIT are associated with:

• Gastrointestinal stromal tumors (GIST)
• Mast cell disease
• Acute myelogenous leukemia
• Piebaldism

Deregulation of KIT receptor tyrosine kinase (TK), particularly the D816V activation loop mutation, is a focal point in therapeutic strategies targeting neoplastic mast cell growth.

Expression and Function:

• Expressed in hematopoietic stem cells, germ cells, mast cells, and gastrointestinal tract Cajal cells.
• Upon binding of stem cell factor (SCF), c-Kit dimerizes, leading to receptor activation and autophosphorylation.
• Tyrosine 703 phosphorylation enables Grb2 docking and Ras/ERK pathway activation.
• SCF/c-Kit signaling activates PI3K, PLC-γ, and JAK/STAT pathways.
• Essential for hematopoiesis, stem cell maintenance, and gametogenesis.
