Target Information
KIT (c-KIT)은 MGF(mast cell growth factor, stem cell factor) 수용체로 작용하는 프로토온코진이자 type 3 transmembrane receptor입니다.
KIT은 원래 feline sarcoma viral oncogene v-kit의 세포 상동체로 확인되었으며, 다양한 조혈세포 및 비조혈세포의 성장과 활성화를 조절합니다.
KIT 유전자의 변이는 위장관 기질종양(GIST), 비만세포질환, 급성골수성백혈병, piebaldism 등과 관련이 있습니다.
특히 활성 루프 변이 D816V는 비정상적인 KIT 수용체의 티로신키나제 활성화를 유도하며, 이는 비만세포 종양 성장 억제 치료 전략의 주요 표적이 됩니다.
c-Kit은 조혈줄기세포, 생식세포, 비만세포, 위장관의 Cajal 세포에서 발현됩니다.
SCF(stem cell factor)와 결합 시, c-Kit은 이합체를 형성하고 자가 인산화를 통해 Grb2 결합 및 Ras/ERK 신호 경로를 활성화합니다.
또한 PI3K, PLC-γ, JAK/STAT 등의 하위 신호 경로를 활성화하며, 조혈, 줄기세포 유지, 생식세포 형성에 필수적인 역할을 합니다.
