Target Information
Cas9 is an RNA-guided DNA endonuclease enzyme associated with the CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) adaptive immunity system in bacteria and genetic engineering.
This system uses RNA to direct the nuclease to specific DNA sequences, enabling targeted genome editing such as indel mutations, sequence insertions, deletions, or rearrangements.
Cas9 can also be used to modulate gene expression, histone modifications, or DNA methylation.
Two common forms are derived from Streptococcus pyogenes (SpCas9) and Staphylococcus aureus (SaCas9). SaCas9 is smaller, efficient, and exhibits high specificity, recognizing NNGRRT PAM sequences and cleaving DNA at comparable rates to SpCas9.
Its compact size makes it ideal for all-in-one delivery systems using adeno-associated vectors.
